The run, which has been organised by the Lions Club, will begin at the DAF Trucks car park on the bypass in Thame. Entrants can run or walk either the 1.5 mile version or three mile version of the route.
Both courses will finish at the Scout building on the recreation ground where the guides will be serving mince pies and mulled wine.
A large entry is expected from the Rainbows, Brownies, and the Guides. Other youth groups in Thame have been invited to take part including the Army Cadets, Air Cadets and Scouts along with Thame Runners, Buddies and the Monday morning running club.
The old Guiding Centre was knocked down recently after 37 years of service, to make way for the modern, new centre.
Nearly 190 girls from Thame attend Rainbows, Brownies or Guides. Aged five to 16, they account for 25 per cent of girls in that age range throughout the town.
Linda Newton, district commissioner for Guides in Thame, said: "The building has been knocked down which is very exciting for us. We should be back in by Easter and we are just short of our £350,000 target."
The run starts at 11am and a free Santa hat and bottle of water will be provided. There will also be a prize for the best dressed Santa. The routes will be fully marshalled using mainly the phoenix trail.

Adult: £3 (OAP £1), Under 16: £1 Family entry: £6
Entries received after December 1, are: Adult: £3.50 (OAP £1), or on the day Under 16: £1.50, Family entry: £6.50
